Developing an original nanopore glycan sequencing technology to foster a new “foundational technology” in life sciences and advance innovative drug research and discovery. As one of the three major biological substances, glycans are crucial entities of biological information and drug molecules. The structural complexity of glycans far exceeds that of nucleic acids and proteins, and glycan sequencing has long been regarded as an “impossible task”. Based on long-term research on ion channels and in-depth understanding of the electrophysiology of biological pores, and building on the development of nucleic acid and protein sequencing technologies, he proposes the concept and idea of glycan sequencing using biological nanopores, map the technical roadmap for nanopore-based glycan sequencing, and complete the proof-of-concept for three technical routes: “hydrolysis sequencing”, “chain sequencing”, and “assembly sequencing”. He has pioneered the sequential analysis of glycan chain structural information at the single-molecule level, leading the foundational technology of nanopore-based glycan sequencing into a new stage.
